Functional analysis of deubiquitylases in Drosophila melanogaster

Reversible ubiquitylation of proteins is a crucial mechanism regulating fundamental cellular processes such as cell proliferation, cell death, gene expression and signal transduction. In this very dynamic process, ubiquitin molecules attached covalently to substrate proteins by ubiquitylating enzymes, and removed by deubiquitylating enzymes, or DUBs. Despite the importance of DUBs and the fact that a number of studies have addressed their enzymatic properties and mode of action in recent years, our knowledge about their physiological importance and regulation is incomplete. In order to determine their physiological importance and to get deeper insight into their cellular function and regulation, we carry out a systematic genetic analysis of different mutant lines, each disrupting a gene coding for one of the DUB enzymes in the genetically well tractable experimental organism, Drosophila melanogaster. Interested PhD student could join us and carry out genetic complementation screens in order to identify DUBs with possible roles in tumorgenezis, protein degradation, stem cell differentiation, spermatogenesis and autophagy. He or she will be involved in more detailed functional analysis of selected DUBs with interesting interaction phenotypes. Since ubiquitylation and deubiquitylation is essential for normal cell division, an aberrant activation or inactivation of these opposing processes can lead to unwanted cell division, which can result in cancer. Misregulation of some of them has already been linked to tumorgenezis. Therefore we hope that our research outlined in this application will not only broaden our knowledge of DUB function and regulation, but it will also empower us to manipulate DUB activities as a new way of therapeutic strategy against cancer.
