Description of the research topic:
The repressive nature of nucleosomal structure predicts nucleosome eviction as a primary mechanism of gene regulation. Since DNA assumes a superhelical structure upon winding around the nuceosomes, changes of superhelical density can also influence the interaction between the histones and the DNA. These mutual interactions will be studied in permeabilized cell systems by confocal microscopy and laser scanning cytometry.
