The euchromatin-heterochromatin dichotomy is a well-known light- and electronmicroscopic correlate of the activity and inactivity of the genome, respectively, generally interpreted in terms of packaging differences that, based on common sense, are expected to reflect or determine chromatin accessibility to regulatory macromolecules. A differential accessibility of the two types of chromatin, however, has never been experimentaly verified. Likewise, no clear correlation with gene activity has been clearly documented for the different levels of chromatin’s structural hierarchy. The only structural feature with unequivocally repressive character is the presence of the nucleosomes on inactive promoters and enhancers opposed to the nucleosome free regulatory regions of the active genes, and the relatively decreased stability of nucleosomes along the transcribed loci. Using a laser scanning cytometric approach recently elaborated in our lab for the analysis of nucleosome eviction in a histone modification and cell-cycle dependent manner in situ, combined with fluorescence resonance energy transfer methodology we set out to fill the above, crucial conceptual gap in our understanding of the main principles of gene regulation in higher eukaryotes involving the human.
