Synthesis and functional characterization of peptide-conjugate with calpain activator/inhibitor effect

Description of the research topic:

"Synthesis of enzyme activator conjugates with conjugation of calpastatin A and C peptide and cell-penetrating peptide will be carried out using different chemical bond. The internalization of conjugates will be studied by flow cytometry, the enzyme activating effect will be examined using fluorescence substrate conjugates. New substrate-conjugate will be developed with conjugation of substrate peptide and cell-penetrating peptide, which is able to sign the activity of intracellular calpain. Calpain activity will be measured with it in live cell by flow cytometry and the optimalisation of measurement will be done.
Development of different peptide based inhibitors, characterization of the compounds, measurement their inhibitor effect and selectivity. Conjugation of the most potent and the most selective inhibitors with cell-penetrating peptides will be carried out and their inhibitor activity and selectivity will be studied."
